چکیده

Fatty acids can be synthesized in mitochondrial membranes of rat liver either by elongation or by synthesis de nouo. The outer membranes incorporate greater amounts of substrate into fatty acids than do the inner membranes. Outer membranes synthesize 6and g-carbon fatty acids completely de nouo, while smaller amounts of lo-carbon acids are produced by the mechanism de novo. The reactions de novo utilize acetyl coenzyme A more effectively than malonyl coenzyme A, and synthesis de novo occurs in the absence of exogenous ATP. Elongation, which produces fatty acids 14 carbon atoms long and longer, depends on added ATP, and acetyl-CoA and malonyl-CoA are incorporated about equally. Synthesis de novo of shorter chain fatty acids by outer membranes continues in the presence of ATP and the concomitant elongation reactions. Inner membranes synthesize only longer fatty acids by elongation. If synthesis de novo of fatty acids occurs in the inner membranes, these acids might be removed by oxidation. The ATP concentrations in these experiments were compared with the levels of ATP produced by actively respiring liver mitochondria. During metabolically adverse conditions within the cell, the capacity for manufacturing fatty acids by elongation may fluctuate with the ATP levels whereas synthesis de nouo continues.

Study of the transfer of phospholipids from the endoplasmic reticulum to the outer and inner mitochondrial membranes.

Isolated mitochondria cannot synthesize their own phospholipids, there is only an exchange between exogenous and mitochondrial phospholipid fatty acids. In vitro, the endoplasmic reticulum phospholipids exchange with the phospholipids of the mitochondrial outer and inner membranes. Exchange of the endoplasmic reticulum phospholipids with those of the inner membrane is the same when the incubati...

Incorporation of fatty acids into the outer and inner membranes of isolated rat liver mitochondria.

Acyl-CoA: phospholipid acyl-transferase activity as well as phospholipase A activity were detected in inner and outer membrane preparations from rat liver mitochondria. Both enzyme systems have an optimum pH around 8 and act preferentially on phosphatidylethanolamine. While phospholipase A activity is much lower in the inner membrane than in the outer membrane of mitochondria the reverse is tru...

Sites of biosynthesis of outer and inner membrane proteins of Neurospora crassa mitochondria.

Outer and inner membranes of Neurospora crassa mitochondria were separated by the combined swelling, shrinking, sonication procedure. Membranes were characterized by electron microscopy and by marker enzyme activities. A red carotenoid pigment was found to be concentrated in the outer membrane. The inner mitochondrial membrane was resolved into about 20 protein bands on polyacrylarnide gel elec...

Plant Mitochondrial Electron Transfer and Molecular Biology.

Aerobic respiration, a process performed by all plants, involves the controlled oxidation of metabolites containing reduced carbon to produce carbon dioxide and water as the final products (Taiz and Zeiger, 1991). Severa1 types of reduced carbon compounds, including fatty acids, organic acids, and amino acids, can serve as the primary reducing substrates for plant respiration. However, the most...
